چکیده

We have examined the question, is the carbohydrate moiety of ovalbumin attached to nascent chains or to the completed protein after its release from polyribosomes. Ovalbumin nascent chains have been isolated from hen oviduct by immunoprecipitation of ovalbumin-synthesizing polysomes and subsequent isolation of peptidyl-tRNA by DEAE-cellulose chromatography. Our results indicate that both glucosamine and mannose are incorporated into ovalbumin nascent chains by hen oviduct fragments. Although the asparagine to which the carbohydrates are attached is 10 amino acid residues from the NH2 terminus, the majority of the carbohydrate, if not all, is attached only to those peptide chains that have been essentially completed.

Isolation of hen oviduct ovalbumin and rat live albumin polysomes by indirect immunoprecipitation.

The polyribosomes synthesizing ovalbumin and albumin have been isolated from total oviduct polysomes and total rat liver polysomes, respectively. The isolation was performed using an indirect immunoprecipitation technique which is based on the specific reaction which occurs between antibody against a purified native protein and the nascent peptide chains on polysomes synthesizing that protein. ...

Enzymatic conversion of proteins to glycoproteins.

The enzymatic transfer of the oligosaccharide moiety from an oligosaccharide-lipid to denatured forms of three secretory proteins--ovalbumin, alpha-lactalbumin, and ribonuclease A--has been demonstrated utilizing a membrane fraction from hen oviduct. Based on a survey of 10 proteins denatured by sulfitolysis, the presence of the tripeptide sequence -Asn-X-Thr-Ser- (X represents a variable amino...

Hen oviduct signal peptidase is an integral membrane protein.

Membrane preparations from rough endoplasmic reticulum of hen oviduct resemble those of dog pancreas in their capacity to translocate nascent secretory proteins into membrane vesicles present during cell-free protein synthesis. As with the dog membranes, the precursor form of human placental lactogen is transported into the vesicles and processed to the native secretory form by an associated "s...

Identification and Isolation of Ovalbumin-synthesizing Polysomes

Polysomes involved in ovalbumin synthesis were identified by the binding of 1251-anti-ovalbumin to hen oviduct polysomes. Techniques were developed for the isolation of undegraded hen oviduct polysomes and for the preparation of 1251-y-globulin free of ribonuclease activity. The distribution of 1251-anti-ovalbumin in the polysome profile is in accordance with the size of the polysomes that are ...

Site-specific de-N-glycosylation of diglycosylated ovalbumin in hen oviduct by endogenous peptide: N-glycanase as a quality control system for newly synthesized proteins.

Hen ovalbumin (OVA) is known to exist as a singly N-glycosylated form with a glycan chain on Asn-292 in egg white. Previous studies showed that di-N-glycosylated form of OVA [Di-OVA; CHO-Asn-292/CHO-Asn-311 (CHO, N-glycan chain)], which has two N-glycan chains on Asn-292 and Asn-311, was expressed only transiently in hen oviduct. Di-OVA was not found in egg white, suggesting that this form cann...
